ACE-031 Myostatin Inhibitor and Muscle Growth Outcomes

From a coaching standpoint, ACE-031 was a powerful but flawed tool. It’s a fusion protein made from ActRIIB and an Fc fragment that blocks myostatin, the hormone that limits muscle growth. Early trials showed rapid results, around 3–5% lean mass gains in just a few weeks and solid strength increases.

But as a coach, results don’t matter if the tool isn’t safe. Serious vascular side effects like bleeding gums and nosebleeds showed up, and by 2011, regulatory bodies shut down the program. Acceleron Pharma discontinued development, but the takeaway for coaches and athletes was clear: muscle growth has to be controlled and sustainable.

That lesson helped drive the push toward safer, second-generation myostatin inhibitors with better risk management.

What ACE-031 Is and How It Blocks Myostatin​

ACE-031 is a new type of therapeutic protein called an ActRIIB-Fc fusion protein. This substance works by attaching to myostatin, a protein that your body makes that controls muscle growth.
